Mariano Que was born into modest circumstances in the Philippines, a country that would soon be thrust into the turmoil of World War II. Orphaned at a young age, Que faced significant challenges early on. His education was cut short, ending at the seventh grade due to financial constraints and the loss of his parents during the Japanese occupation. Despite these setbacks, Que's formative years instilled in him a strong work ethic and a compassionate spirit, qualities that would define his later success.

Before the war, Que worked as an employee at Farmacia Central, a prominent pharmacy in Manila owned by Jose Tee Han Kee. This early exposure to the pharmaceutical trade gave him invaluable knowledge about medicines and their importance to public health. However, the devastation of World War II left Manila in ruins, destroying businesses, including the drugstore where Que worked, and leaving the population in dire need of basic necessities like medicine.
